    Jinyu Chen1, Long Shi1,*, Qisheng Huang2, Taotao Wang3, Daojing He4

    CMES-Computer Modeling in Engineering & Sciences, Vol.139, No.2, pp. 1755-1773, 2024, DOI:10.32604/cmes.2023.046826

Abstract In permissioned blockchain networks, the Proof of Authority (PoA) consensus, which uses the election of authorized nodes to validate transactions and blocks, has been widely advocated thanks to its high transaction throughput and fault tolerance. However, PoA suffers from the drawback of centralization dominated by a limited number of authorized nodes and the lack of anonymity due to the round-robin block proposal mechanism. As a result, traditional PoA is vulnerable to a single point of failure that compromises the security of the blockchain network. To address these issues, we propose a novel decentralized reputation management mechanism for permissioned blockchain networks… More > Graphic Abstract

Abstract Gastric cancers are caused primarily due to the activation and amplification of the EGFR or HER2 kinases resulting in cell proliferation, adhesion, angiogenesis, and metastasis. Conventional therapies are ineffective due to the intra-tumoral heterogeneity and concomitant genetic mutations. Hence, dual inhibition strategies are recommended to increase potency and reduce cytotoxicity. In this study, we have conducted computational high-throughput screening of the ChemBridge library followed by in vitro assays and identified novel selective inhibitors that have a dual impediment of EGFR/HER2 kinase activities. Diversity-based High-throughput Virtual Screening (D-HTVS) was used to screen the whole ChemBridge small molecular library against EGFR and… More > Graphic Abstract

Abstract Familial adenomatous polyposis (FAP) is an autosomal dominant disease with a high probability of becoming cancerous. Many RNAs potentially associated with FAP have not been identified. In this study, a circRNA (circular RNA) expression profile of FAP was established using a circRNA microarray, and differentially expressed circRNAs were verified by RT-qPCR. The effects of hsa_circ_0036740 on the malignant behavior of tumor cells (proliferation, apoptosis, and epithelial mesenchymal transition) and the levels of C3A complement protein expression were evaluated. Moreover, neutrophils were isolated and co-cultured with colorectal cancer cells (CRCs), followed by measurements of MPO-DNA, citrullinated histone H3, interleukin (IL)-1β, IL-6,… More > Graphic Abstract

    Abstract

Myc belongs to a family of proto-oncogenes that encode transcription factors. The overexpression of c-Myc causes many types of cancers. Recently, we established a system for screening c-Myc inhibitors and identified antimycin A by screening the RIKEN NPDepo chemical library. The specific mechanism of promoting tumor cell metastasis by high c-Myc expression remains to be explained. In this study, we screened approximately 5,600 microbial extracts using this system and identified a broth prepared from Streptomyces sp. RK19-A0402 strongly inhibits c-Myc transcriptional activity. After purification of the hit broth, we identified compounds closely related to the aglycone of cytovaricin and had… More > Graphic Abstract

Abstract β-transducin repeat-containing protein (β-TrCP) is an F-box protein subunit of the E3 Skp1-Cullin-F box (SCF) type ubiquitin-ligase complex, and provides the substrate specificity for the ligase. To find potent ligands of β-TrCP useful for the proteolysis targeting chimera (PROTAC) system using β-TrCP in the future, we developed a high-throughput screening system for small molecule β-TrCP ligands. We screened the chemical library utilizing the system and obtained several hit compounds. The effects of the hit compounds on in vitro ubiquitination activity of SCFβ-TrCP1 and on downstream signaling pathways were examined. Hit compounds NPD5943, NPL62020-01, and NPL42040-01 inhibited the TNFα-induced degradation of… More > Graphic Abstract

The primary objective of database and application developers is to provide scalable and reliable database systems. This research presents a novel method for data partitioning and load balancing for scalable transactions. Data is efficiently partitioned using the hybrid graph partitioning method. Optimized load balancing (OLB) approach is applied to calculate the weight factor, average workload, and partition efficiency. The presented approach is appropriate for various online data transaction applications. The quality of the proposed approach is examined using OLTP database benchmark. The performance of the… More >

    Computer Systems Science and Engineering, Vol.46, No.1, pp. 73-92, 2023, DOI:10.32604/csse.2023.034461

Abstract Increasing the coverage and capacity of cellular networks by deploying additional base stations is one of the fundamental objectives of fifth-generation (5G) networks. However, it leads to performance degradation and huge spectral consumption due to the massive densification of connected devices and simultaneous access demand. To meet these access conditions and improve Quality of Service, resource allocation (RA) should be carefully optimized. Traditionally, RA problems are nonconvex optimizations, which are performed using heuristic methods, such as genetic algorithm, particle swarm optimization, and simulated annealing. However, the application of these approaches remains computationally expensive and unattractive for dense cellular networks. Therefore,… More >

Abstract Background and Aim: Hundreds of consistently altered metabolic genes have been identified in breast cancer (BC), but their prognostic value remains to be explored. Therefore, we aimed to build a prediction model based on metabolism-related genes (MRGs) to guide BC prognosis. Methods: Current work focuses on constructing a novel MRGs signature to predict the prognosis of BC patients using MRGs derived from the Virtual Metabolic Human (VMH) database, and expression profiles and clinicopathological data from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) databases. Results: The 3-MRGs-signature constructed by SERPINA1, QPRT and PXDNL was found to be an… More >

Reconfigurable Sensing Time in Cooperative Cognitive Network Using Machine Learning

    Noor Gul1,2, Saeed Ahmed1,3, Su Min Kim1, Muhammad Sajjad Khan4, Junsu Kim1,*

    CMC-Computers, Materials & Continua, Vol.74, No.3, pp. 5209-5227, 2023, DOI:10.32604/cmc.2023.026945

Abstract A cognitive radio network (CRN) intelligently utilizes the available spectral resources by sensing and learning from the radio environment to maximize spectrum utilization. In CRNs, the secondary users (SUs) opportunistically access the primary users (PUs) spectrum. Therefore, unambiguous detection of the PU channel occupancy is the most critical aspect of the operations of CRNs. Cooperative spectrum sensing (CSS) is rated as the best choice for making reliable sensing decisions. This paper employs machine-learning tools to sense the PU channels reliably in CSS. The sensing parameters are reconfigured to maximize the spectrum utilization while reducing sensing error and cost with improved… More >

Abstract The simultaneous wireless information and power transfer (SWIPT) relay system is one of the emerging technologies. Xiaomi Corporation and Motorola Inc. recently launched indoor wireless power transfer equipment is one of the most promising applications. To tap the potential of the system, hybrid automatic repeat request (HARQ) is introduced into the SWIPT relay system. Firstly, the time slot structure of HARQ scheme based on full duplex two-way amplify and forward (AF) SWIPT relay is given, and its retransmission status is analyzed. Secondly, the equivalent signal-to-noise ratio and outage probability of various states are calculated by approximate simplification. Thirdly, the energy… More >
