Abstract The calla lily (Zantedeschia spreng.) is a bulbous flower native to the tropical regions of Africa. Calla lily has gained significant popularity in the international market owing to its intricate morphology and prolonged flowering duration. Despite such advantages, for two sub-groups of calla lily, known as group Zantedeschia and group Aestivae, there are challenges in terms of hybrid production due to the ‘plastome-genome incompatibility’ therebetween. Tissue culture is a fundamental biotechnological tool used in gene editing research, with a focus on disease resistance and flower color in calla lily breeding programs. The present review provides a brief… More >

    Chunxue Peng1,2, Fang Gao1,2, Hao Wang1,2, Iraida Nikolaevna Tretyakova3, Alexander Mikhaylovich Nosov4,5, Hailong Shen1,2,*, Ling Yang1,2,*

    Phyton-International Journal of Experimental Botany, Vol.91, No.1, pp. 223-238, 2022, DOI:10.32604/phyton.2022.015523 - 16 August 2021

Abstract Korean pine is an important afforestation tree species in Northeast China, which has a high ecological and economic value. Although regeneration of somatic embryogenesis using immature zygotic embryos of Korean pine as explants has been successful, it cannot be applied to automation and large-scale production. Therefore, we urgently need a method that can increase the output of somatic embryos (SEs) to meet the needs of large-scale production. We used Korean pine 1–1 and 1–100 cell lines as research materials to evaluate the effects of inoculum-density, culture time, orbiting speed, vessel volume, plant growth regulator (PGR)… More >

The Regulatory Roles of microRNAs and Associated Target Genes during Early Somatic Embryogenesis in Liriodendron Sino-Americanum

    Dan Wang1, Fengjuan Lu1, Pengkai Wang1, Jiaji Zhang1, Lu Lu1, Yan Zhen1, Ye Lu1, Tielong Cheng2, Jisen Shi1,*, Jinhui Chen1,*

    Phyton-International Journal of Experimental Botany, Vol.90, No.5, pp. 1445-1463, 2021, DOI:10.32604/phyton.2021.015499 - 27 May 2021

Abstract Somatic cells respond to considerable stress, and go through a series of phytohormone pathways, then forming an embryo. The developmental process is recorded as somatic embryogenesis (SE). One of the key components regulating SE are the microRNAs (miRNAs). Despite previous studies, it is still not clear exactly how miRNAs exert their function of regulating targets during conditionally activated early SE. Here, we use Liriodendron sino-americanum as a model system and perform a combined analysis of microfluidic chips and degradome sequencing to study this process. We identified a total of 386 conserved miRNAs and 153 novel miRNAs… More >

    Phyton-International Journal of Experimental Botany, Vol.89, No.4, pp. 779-794, 2020, DOI:10.32604/phyton.2020.09421 - 09 November 2020

Abstract The use of medicinal plants for different therapeutic values is well documented in African continent. African diverse biodiversity hotspots provide a wide range of endemic species, which ensures a potential medicinal value. The feasible conservation approach and sustainable harvesting for the medicinal species remains a huge challenge. However, conservation approach through different biotechnological tools such as micropropagation, somatic embryogenesis, synthetic seed production, hairy root culture, molecular markers based study and cryopreservation of endemic African medicinal species is much crucial. In this review, an attempt has been made to provide different in vitro biotechnological approaches for the More >

Exogenous Spermidine Promotes Somatic Embryogenesis of Cunninghamia lanceolata by Altering the Endogenous Phytohormone Content

    Dandan Wang1,#, Yulin Guo1,#, Xiaofei Long1, Yan Pan1, Dingjie Yang1, Rui Li1, Ye Lu1, Ying Chen1,2, Jisen Shi1, Jinhui Chen1,*

    Phyton-International Journal of Experimental Botany, Vol.89, No.1, pp. 27-34, 2020, DOI:10.32604/phyton.2020.08971 - 01 March 2020

Abstract In order to study how exogenous hormones in C. lanceolata (gymnosperm) regulate somatic embryogenesis, we measured the endogenous phytohormones of two genotypes with different somatic embryogenesis efficiency and found that an increase in endogenous concentrations of IAA and ABA may be correlated to more efficient somatic embryogenesis. By applying exogenous spermidine, we found that exogenous hormones may affect somatic embryogenesis efficiency through affecting the endogenous phytohormone content. Based on these results, further studies can be conducted whereby the concentration of exogenous hormones or the levels of endogenous phytohormones by molecular methods are regulated to promote somatic More >

    Menéndez-Yuffá1 A, L Ríos-Bolívar2

    Phyton-International Journal of Experimental Botany, Vol.77, pp. 49-64, 2008, DOI:10.32604/phyton.2008.77.049

Abstract Regeneration and multiplication of coffee (Coffea arabica L.) through somatic embryogenesis can be efficiently achieved through different methods. However, it is very important to analyze the progenies obtained by these methods in order to establish their genetic stability. The objectives of this research were (1) to determine the protein electrophoretic patterns in leaves of coffee vitroplants which were regenerated through somatic embryogenesis, and (2) to compare these patterns with those of the explant donor plants. The protein patterns of some coffee genotypes were initially determined to detect possible differences among them. Each analyzed genotype showed a More >
