CAF-derived exosome-miR-3124-5p promotes malignant biological processes in NSCLC via the TOLLIP/TLR4-MyD88-NF-κB pathway

    TAO SUN1,2, QINGHUA SONG3, HUA LIU1,*

    Oncology Research, Vol.33, No.1, pp. 133-148, 2025, DOI:10.32604/or.2024.054141 - 20 December 2024

Abstract Background: Lung cancer is a life-threatening disease that occurs worldwide, but is especially common in China. The crucial role of the tumour microenvironment (TME) in non-small cell lung cancer (NSCLC) has attracted recent attention. Cancer-associated fibroblasts (CAFs) are the main factors that contribute to the TME function, and CAF exosomes are closely linked to NSCLC. Methods: The expression levels of miR-3124-5p and Toll-interacting protein (TOLLIP) were analysed by bioinformatics prediction combined with RT-qPCR/Western Blot detection. Fibroblasts were isolated and identified from clinical NSCLC tissues. Transmission electron microscopy and Western Blot were used to identify exosomes… More >

Huiru Deng#, Huan Tian#, Liuyan Yang, Siyang Ou, Hui Wang, Guangdong Geng*, Suqin Zhang*

    Phyton-International Journal of Experimental Botany, Vol.93, No.12, pp. 3273-3291, 2024, DOI:10.32604/phyton.2024.058638 - 31 December 2024

Abstract Hot pepper (Capsicum annuum var. conoides) is a significant vegetable that is widely cultivated around the world. Currently, global climate change has caused frequent severe weather events, and waterlogging stress harms the pepper industry by affecting the planting period, growth conditions, and disease susceptibility. The gene CaABI3/VP1-1 could improve pepper waterlogging tolerance. In order to explore the upstream regulatory mechanism of CaABI3/VP1-1, a high-quality standardized yeast hybrid library was successfully constructed for yeast one-, two-, and three-hybrid screening using pepper ‘ZHC2’ as the experimental material, with a library recombinant efficiency of up to 100%. The length of inserted… More >

Abstract Background: Inhibitor of NF-κB kinase-interacting protein (IKIP) is known to promote proliferation of glioblastoma (GBM) cells, but how it affects migration and invasion by those cells is unclear. Methods: We compared levels of IKIP between glioma tissues and normal brain tissue in clinical samples and public databases. We examined the effects of IKIP overexpression and knockdown on the migration and invasion of GBM using transwell and wound healing assays, and we compared the transcriptomes under these different conditions to identify the molecular mechanisms involved. Results: Based on data from our clinical samples and from public databases, More >

A Brachypodium distachyon calcineurin B-like protein-interacting protein kinase, BdCIPK26, enhances plant adaption to drought and high salinity stress

    QINGCHEN LUO1,#,*, JIALU FENG2,#, XIUQI DENG1

    BIOCELL, Vol.47, No.5, pp. 1145-1158, 2023, DOI:10.32604/biocell.2023.027847 - 10 April 2023

Abstract As sessile organisms, plants possess a complex system to cope with environmental changes. Ca2+ functions as a vital second messenger in the stress signaling of plants, and the CBL-interacting protein kinases (CIPKs) serve as essential elements in the plant Ca2+ signaling pathway. In this study, calcineurin B-like protein-interacting protein kinase 26 (BdCIPK26) from Brachypodium distachyon was characterized. Overexpression of BdCIPK26 enhanced tolerance to drought and salt stress of transgenic plants. Further investigations revealed that BdCIPK26 participated in abscisic acid (ABA) signaling, conferred hypersensitivity to exogenous ABA in transgenic plants, and promoted endogenous ABA biosynthesis. Moreover, BdCIPK26 was found More >

Genome Wide Characterization of CBL-CIPK Family Genes and Their Responsive Expression in Rosa chinensis

Abstract Calcium (Ca2+) plays a pivotal role in various signal transduction pathways. Calcineurin B-like proteins (CBLs) are a unique group of Ca2+ sensors that decode Ca2+ signals by activating the plant specific protein kinase known as the CBL-interacting protein kinase (CIPK). In plants, the CBL-CIPK signaling network regulates multiple signals in response to different extracellular cues including abiotic stress. However, the genome wide annotation and expression patterns of CBLs and CIPKs in woody cutting flower plants are still unclear. In this study, a total number of 7 CBLs (RcCBLs) and 17 CIPKs (RcCIPKs) genes, divided into four and five subfamilies,… More >

Cylindromatosis Is Required for Survival of a Subset of Melanoma

Ting La*†, Lei Jin*‡, Xiao Ying Liu*, Ze Hua Song*, Margaret Farrelly, Yu Chen Feng, Xu Guang Yan, Yuan Yuan Zhang*,† Rick F. Thorne*,*§ Xu Dong Zhang*†, Liu Teng*

Abstract The deubiquitinase cylindromatosis (CYLD) functions as a tumor suppressor inhibiting cell proliferation in many cancer types including melanoma. Here we present evidence that a proportion of melanoma cells are nonetheless addicted to CYLD for survival. The expression levels of CYLD varied widely in melanoma cell lines and melanomas in vivo, with a subset of melanoma cell lines and melanomas displaying even higher levels of CYLD than melanocyte lines and nevi, respectively. Strikingly, although short hairpin RNA (shRNA) knockdown of CYLD promoted, as anticipated, cell proliferation in some melanoma cell lines, it reduced cell viability in… More >

Novel BRCA2-Interacting Protein, LIMD1, Is Essential for the Centrosome Localization of BRCA2 in Esophageal Cancer Cell

Xiaobin Hou*1, Tinghui Li†1, Zhipeng Ren*, Yang Liu*

    Oncology Research, Vol.24, No.4, pp. 247-253, 2016, DOI:10.3727/096504016X14652175055765

Abstract Mutation of breast cancer 2, early onset (BRCA2) has been identified as a vital risk factor for esophageal cancer (EC). To date, several proteins have been reported as BRCA2-interacting proteins and are associated with multiple biological processes. This study's aim was to identify a novel interactive protein of BRCA2 and to explore its functional roles in EC. A yeast two-hybrid screening was performed to identify a novel BRCA2-interacting protein. Glutathione-S-transferase (GST) pull-down analysis was performed to find out how the binding domain of BRCA2 interacts with LIM domains containing 1 (LIMD1). The interaction between LIMD1… More >

Knockdown of HPIP Inhibits the Proliferation and Invasion of Head-and-Neck Squamous Cell Carcinoma Cells by Regulating PI3K/Akt Signaling Pathway

Yangjing Chen, Ruimin Zhao, Qian Zhao, Yuan Shao, Shaoqiang Zhang

    Oncology Research, Vol.24, No.3, pp. 153-160, 2016, DOI:10.3727/096504016X14612603423476

Abstract Hematopoietic pre-B-cell leukemia transcription factor (PBX)-interacting protein (HPIP/PBXIP1) is a corepressor for the transcription factor PBX. Previous studies showed that HPIP is frequently overexpressed in many tumors. However, the role of HPIP in head-and-neck squamous cell carcinoma (HNSCC) has not yet been determined. Thus, we decided to investigate the effects and mechanisms of HPIP in HNSCC. Our results demonstrated that HPIP is highly expressed in human HNSCC cell lines and provides the first evidence that knockdown of HPIP obviously inhibits proliferation and migration/invasion in HNSCC cells in vitro, as well as inhibits tumor growth in More >
