Non-canonical BRAF variants and rearrangements in hairy cell leukemia

    STEPHEN E. LANGABEER*

    Oncology Research, Vol.32, No.9, pp. 1423-1427, 2024, DOI:10.32604/or.2024.051218 - 23 August 2024

Abstract Hairy cell leukemia (HCL) is an uncommon mature B-cell malignancy characterized by a typical morphology, immunophenotype, and clinical profile. The vast majority of HCL patients harbor the canonical BRAF V600E mutation which has become a rationalized target of the subsequently deregulated RAS-RAF-MEK-MAPK signaling pathway in HCL patients who have relapsed or who are refractory to front-line therapy. However, several HCL patients with a classical phenotype display non-canonical BRAF mutations or rearrangements. These include sequence variants within alternative exons and an oncogenic fusion with the IGH gene. Care must be taken in the molecular diagnostic work-up of patients More >

Abstract Background: Cochlear hair cell injury is a common pathological feature of hearing loss. The basic helix-loop-helix family, member e40 (Bhlhe40), a gene belonging to the basic helix-loop-helix (bHLH) family, exhibits strong transcriptional repression activity. Methods: Oxidative damage, in House Ear Institute-Organ of Corti 1 (HEI‑OC1) cells, was caused using hydrogen peroxide (HO). The Ad-Bhlhe40 particles were constructed to overexpress Bhlhe40 in HEI-OC1 cells. Various assays including cell counting kit-8 (CCK-8), terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling assay (TUNEL), flow cytometry, immunofluorescence, and corresponding commercial kits were employed to investigate the impacts of Bhlhe40 on cell viability, apoptosis,… More > Graphic Abstract

Abstract Panax ginseng C. A. Mey. is an important plant species used in traditional Chinese medicine, whose primary active ingredient is a ginsenoside. Ginsenoside biosynthesis is not only regulated by transcription factors but also controlled by a variety of structural genes. Nonetheless, the molecular mechanism underlying ginsenoside biosynthesis has always been a topic in the discussion of ginseng secondary metabolites. Squalene epoxidase (SQE) is a key enzyme in the mevalonic acid pathway, which affects the biosynthesis of secondary metabolites such as terpenoid. Using ginseng transcriptome, expression, and ginsenoside content databases, this study employed bioinformatic methods to systematically… More >
