Genome-wide analysis of microRNAs induced and regulated by colchicine in mulberry

    MENGMENG WU1,#, PENG GUO1,#, YISU SHI1,#, DANYAN ZHENG1, QIAONAN ZHANG1, XIN JIN1, YUHUA WANG1, QIANG LIN2, RONGJUN FANG1,*, WEIGUO ZHAO1,*

Abstract A variety of plants were colchicine treated to double their chromosome number. Chromosomes are genetic carriers that determine the individual traits of organisms. The doubling of chromosomes will lead to modifications in plant morphology, physiology and genetics. To determine the response of mulberry trees induced by colchicine, using mulberry variety Yu-711 leaves as research materials, two small RNA libraries (control and experimental groups) were constructed. It was found that 45 known miRNA genes and 78 predicted novel miRNA genes in the sequence results. A comparison of data between the control group and the experimental group… More >

Cytogenetical Changes among Polyembryonic (PEm) and Non-PEm Maize Plants

    A. A. Román-Calzoncit1, F. Ramírez-Godina2,*, J. Sánchez-Laureano2, A. C. Flores-Gallegos1, J. Espinoza-Velázquez2, R. Rodríguez-Herrera1,*

    Phyton-International Journal of Experimental Botany, Vol.90, No.1, pp. 87-97, 2021, DOI:10.32604/phyton.2020.013880 - 20 November 2020

Abstract Polyembryony in maize (PEm) contributes to improving the nutritional properties of the grain, as well as an increase in yield, since it generates multiple plants per seed, opening the possibility of developing new varieties. However, it is unknown whether polyembryony in maize is the product of chromosomal abnormalities. Based on the above, in this research a cytogenetic study was proposed to verify if chromosomal abnormalities are related to the maize polyembryony. For a meiotic study, maize genotypes with variable proportions of polyembryony (PEm), from the UA-IMM-BAP population and non-PEm (monoembryonic) maize were used, while for… More >

Study of the mitotic and meiotic chromosomes of sotol (Dasylirion cedrosanum Trel.)

    Hernández-Quintero JD1, MH Reyes-Valdés2, DV Mendoza-Rodríguez3, M Gómez-Martínez2, R Rodríguez-Herrera4

    Phyton-International Journal of Experimental Botany, Vol.84, No.1, pp. 107-112, 2015, DOI:10.32604/phyton.2015.84.107

Abstract The genus Dasylirion is a group of plants typically present in the Chihuahuan Desert, perennial, with a dioecious sexual behavior and commonly called sotoles. This genus has been little studied from the biological point of view, and the bases of its reproductive response remain unknown. In this work we studied the chromosome number and meiotic response of Dasylirion cedrosanum in the county of Saltillo, Coahuila, located at the North East of Mexico. For the preparation of mitotic chromosomes, we used a technique based on enzymatic treatment with pectolyase and cellulase, as well as staining with acetocarmin… More >

Detection of single copy sequences using BAC-FISH and C-PRINS techniques in sunflower chromosomes

    PAOLA TALIA1 , EDUARDO J. GREIZERSTEIN1 , H. ESTEBAN HOPP1,2 , NORMA PANIEGO1 , LIDIA POGGIO2 AND RUTH A. HEINZ1,2*

    BIOCELL, Vol.35, No.1, pp. 19-28, 2011, DOI:10.32604/biocell.2011.35.019

Abstract Bacterial artificial chromosome - fluorescence in situ hybridization (BAC-FISH) and cyclingprimed in situ labeling (C-PRINS) techniques were evaluated for integration of physical and genetic maps of sunflower (Helianthus annuus L.). Single-site SSR markers were selected from three linkage groups of a high-density sunflower genetic map. This selection was based on previously identified QTL associated to S. sclerotiorum. These markers were used to select BACs contaning single copy sequences for BAC-FISH aplication. Blocking of highly dispersed repetitive sunflower sequences reduced unspecific hybridization, and allowed the detection of specific signals for BACs containing SSR markers HA4222 and HA2600, anchored to LG More >

Synaptonemal complexes and XY behavior in two species of argentinian armadillos: Chaetophractus villosus and Dasypus hybridus (Xenarthra, Dasypodidae)

    R. B. SCIURANO, M.S. MERANI, JIMENA BUSTOS, A.J. SOLARI

    BIOCELL, Vol.30, No.1, pp. 57-66, 2006, DOI:10.32604/biocell.2006.30.057

Abstract Spermatocytes from the two armadillo species, C. villosus and D. hybridus were studied in microspreads for synaptonemal complexes (SCs) and in thin sections for electron microscopy (EM). The complete SC karyotype generally agrees with previous reports on mitotic chromosomes, except for the sex chromosomes. The X chromosome is submetacentric in both species and the Y is the shortest one in C. villosus and the second shortest in D. hybridus, and an extremely acrocentric one. A SC is formed along the total length of the Y chromosome, and this SC persists along all the pachytene substages. A single recombination nodule… More >

Visualization of the ribosomal DNA (45S rDNA) of Indica rice with FISH on some phases of cell cycle and extended DNA fibers

    ZONG-YUN LI1,2*, MEI-LI FU1, FANG-FANG HU1, SHU-FENG HUANG1, YUN-CHUN SONG2

    BIOCELL, Vol.30, No.1, pp. 27-32, 2006, DOI:10.32604/biocell.2006.30.027

Abstract The ribosomal DNA (45S rDNA) behaviors during the cell cycle were analyzed on interphase nuclei, prophases, metaphases, pachytene chromosomes and extended DNA fibers in rice (Oryza, sativa ssp.indica cv.Guangluai No.4) by using high-resolution fluorescent in situ hybridization (FISH). The results show that 45S rDNA is located at the ends of short arms of chromosomes 9 and 10. But the signals are much more intense on chromosome 9 than on chromosome 10 in metaphase. Pachytene chromosome has rDNA signal arrays on chromosome 9. Different phases are described and discussed. These results indicate that the activity of rDNA at individual More >

Karyotype description of Pomacea patula catemacensis (Caenogastropoda, Ampullariidae), with an assessment of the taxonomic status of Pomacea patula

    MARÍA ESTHER DIUPOTEX-CHONG1, NÉSTOR J. CAZZANIGA2, ALEJANDRA HERNÁNDEZ-SANTOYO3, JOSÉ MIGUEL BETANCOURT-RULE4

    BIOCELL, Vol.28, No.3, pp. 279-285, 2004, DOI:10.32604/biocell.2004.28.279

Abstract Mitotic chromosomes of the freshwater snail Pomacea patula catemacensis (Baker 1922) were analyzed on gill tissue of specimens from the type locality (Lake Catemaco, Mexico). The diploid number of chromosomes is 2n = 26, including nine metacentric and four submetacentric pairs; therefore, the fundamental number is FN = 52. No sex chromosomes could be identified. The same chromosome number and morphology were already reported for P. flagellata, i.e., the other species of the genus living in Mexico. The basic haploid number for family Ampullariidae was reported to be n° =°14 in the literature; so, its reduction to More >
