Abstract Background: Nasopharyngeal carcinoma (NPC) exhibits a significant prevalence in the southern regions of China, and paclitaxel (PTX) is frequently employed as a medication for managing advanced NPC. However, drug resistance is typically accompanied by a poor prognosis. Exploring the synergistic potential of combining multiple chemotherapeutic agents may represent a promising avenue for optimizing treatment efficacy. Methods: This study investigated whether 3-Methyladenine (3-MA) could potentiated the effect of PTX and its potential molecular mechanism. Samples were divided into the following categories: Negative control (NC) with the solvent dimethyl sulfoxide (DMSO, 0.5% v/v), PTX (400 nM), 3-MA… More >

Abstract Background: Nasopharyngeal carcinoma (NPC) is one of the most prevalent cancers in Southeast Asia. Sirtuin 2 (SIRT2) is a member of the NAD+-dependent deacetylase family and has been shown to play important roles in numerous biological processes. However, Its function in NPC remains uncertain. The primary aim of this study is to clarify the role of SIRT2 in NPC. Methods: In this research, we examined the effect of SIRT2 silencing on NPC cell proliferation and colony formation using vitro NPC cell lines. Co-immunoprecipitation and mass spectrometry was applied to identify SIRT2-interacting proteins in NPC cells. Results:More > Graphic Abstract

Abstract Background: Kinesin family member 15 (KIF15) is a protein that regulates cell mitosis and plays an important role in the development and progression of several types of human cancers. However, the role of KIF15 in the development of nasopharyngeal cancer (NPC) is still unclear. Methods: The differential expression of KIF15 in NPC and para-carcinoma tissues was evaluated based on data collected from Gene Expression Omnibus (GEO) database and immunohistochemical analysis of clinical specimens collected from a patient cohort. Cell lines 5-8F and CNE-2Z were selected for the construction of KIF15‑knockdown cell models. CCK8 assay, flow cytometry,… More >

Abstract Nasopharyngeal carcinoma (NPC) is the most prevalent human primary malignancy of the head and neck, and the presence of vasculogenic mimicry (VM) renders anti-angiogenic therapy ineffective and poorly prognostic. However, the underlying mechanisms are unclear. In the present study, we used miR-940 silencing and overexpression for in vitro NPC cell EdU staining, wound healing assay and 3D cell culture assay, and in vivo xenograft mouse model and VM formation to assess miR-940 function. We found that ectopic miR-940 expression reduced NPC cell proliferation, migration and VM, as well as tumorigenesis in vivo. By bioinformatic analysis, circMAN1A2 was… More >

Icotinib Enhanced Radiosensitization of Nasopharyngeal Carcinoma by Inhibiting the Expression of Epidermal Growth Factor Receptor

    Bo Wu1,#, Huacai Xiong2,#, Yong Wang1, Shankun Zhao1, Hongsheng Lu1, Wei Hu3,*

    Oncologie, Vol.24, No.3, pp. 553-563, 2022, DOI:10.32604/oncologie.2022.023025 - 19 September 2022

Abstract Epidermal growth factor receptor (EGFR) is frequently overexpressed in multiple malignancies. Icotinib (IH), a new EGFR tyrosine kinase inhibitor, enhances radiosensitivity in various types of cancer, but its effect on nasopharyngeal carcinoma (NPC) remains unclear. Total 115 NPC tissue sections and 30 nasopharyngitis tissue sections were enrolled. The correlation of EGFR expression and clinicopathologic features of NPC was analyzed. Survival analysis was calculated by using univariate and multivariate regression analysis. A radioresistant NPC cell line, CNE-2R, was established with a gradient irradiation schedule. Cell viability, colony formation and EGFR expression of CNE-2/2R cells were examined.… More >

Abstract Our previous studies suggested a potential interaction between the POK erythroid myeloid ontogenic factor ZBTB7A and glucose transporter 1 (GLUT1) in nasopharyngeal carcinoma (NPC). This study was designed to confirm the interaction and further evaluate the precise mechanism by which ZBTB7A and GLUT1 regulate NPC development. The binding sites between ZBTB7A and the promoter of GLUT1 were predicted by bioinformatics. Gene expression was measured by quantitative real-time polymerase chain reaction (qPCR), western blotting, and immunohistochemistry. The activities of key glycolysis enzymes, including hexokinase (HK), pyruvate kinase (PK), lactate dehydrogenase (LDH), and lactate, were detected using… More >

Abstract Nasopharyngeal carcinoma (NPC) is one of the most common malignant tumors of the head and neck, and its incidence is the highest all around the world. Intensive radiotherapy using computer-aided diagnosis is the best technique for the treatment of NPC. The key step of radiotherapy is the delineation of the target areas and organs at risk, that is, tumor images segmentation. We proposed the segmentation method of NPC image based on multi-scale cascaded fully convolutional network. It used cascaded network and multi-scale feature for a coarse-to-fine segmentation to improve the segmentation effect. In coarse segmentation,… More >

Abstract The aim of this study was to explore the predictive role of pretreatment MRI-based radiomics on early response of neoadjuvant chemotherapy (NAC) in locoregionally advanced nasopharyngeal carcinoma (NPC) patients. Between January 2016 and December 2016, a total of 108 newly diagnosed NPC patients who were hospitalized in the Cancer Hospital of the University of Chinese Academy of Sciences were reviewed. All patients had complete data of enhanced MR of nasopharynx before treatment, and then received two to three cycles of TP-based NAC. After 2 cycles of NAC, enhanced MR of nasopharynx was conducted again. Compared… More >

PPARb/d Agonist GW501516 Inhibits Tumorigenesis and Promotes Apoptosis of the Undifferentiated Nasopharyngeal Carcinoma C666-1 Cells by Regulating miR-206

Linglan Gu, Yi Shi, Weimin Xu, Yangyang Ji

    Oncology Research, Vol.27, No.8, pp. 923-933, 2019, DOI:10.3727/096504019X15518706875814

Abstract In previous investigations, we reported that peroxisome proliferator-activated receptor / (PPAR / ) activation by GW501516 inhibits proliferation and promotes apoptosis in the undifferentiated C666-1 nasopharyngeal carcinoma (NPC) cells by modulating caspase-dependent apoptotic pathway. In the present study, the mechanism by which GW501516 induces apoptosis was explored from the perspective of microRNA (miRNA) expression. Among the assayed miRNAs that were involved in regulating the expression of antiapoptotic protein Bcl-2, miR-206 was increased significantly and specifically by GW501516 in C666-1 cells at both the in vitro level and at the in vivo xenograft samples. The induction… More >
