Abstract Background: The specific impact of sphingolipid metabolism on developing hepatocellular Carcinoma (HCC) remains unclear. This study aims to explore the relationship between sphingolipid metabolism and HCC prognosis, immune response, and drug sensitivity. Methods: Data were obtained from The Cancer Genome Atlas (TCGA)-Hepatocellular Carcinoma (LIHC) and Gene Expression Omnibus (GEO, GSE14520 datasets). 47 sphingolipid metabolism genes were obtained from the Kyoto Encyclopedia of Genes and Genomes (KEGG) database. After classifying HCC samples using the Non-negative Matrix Factorization (NMF) clustering method, differentially expressed genes were screened. Then, 8 risk genes were obtained by univariate analysis, survival random forest… More > Graphic Abstract

Abstract Background: The regulatory genes related to lipid metabolism affect the intramuscular fat (IMF) content and improve meat quality traits. Mediator Complex Subunit 4 (MED4), a vitamin D receptor protein, affects vitamin D, livestock growth, carcass traits, and triglyceride deposition. However, the physiological function of the MED4 gene on bovine adipocyte differentiation remains unknown. Methods: This study explored the function of the MED4 gene in preadipocyte differentiation in Chinese Red Steppe cattle. The overexpression plasmid and the interference sequences of the MED4 gene were constructed to detect the effects of the MED4 gene on adipogenesis and biomakers using More >

Abstract Lipid is a key component of plasma membrane, which plays an important role in the regulation of various cell biological behaviors, including cell proliferation, growth, differentiation and intracellular signal transduction. Studies have shown that abnormal lipid metabolism is involved in many malignant processes, including colorectal cancer (CRC). Lipid metabolism in CRC cells can be regulated not only by intracellular signals, but also by various components in the tumor microenvironment, including various cells, cytokines, DNA, RNA, and nutrients including lipids. In contrast, abnormal lipid metabolism provides energy and nutrition support for abnormal malignant growth and distal More >

Abstract In order to explore the role of forkhead box protein O1 (FoxO1) in the lipid metabolism and cell proliferation, goose primary hepatocytes were isolated and incubated with insulin or PI3K-Akt-mTOR pathway dual inhibitor NVP-BEZ235, and then transfected with FoxO1 interference plasmid. The related parameters of lipid metabolism and cell proliferation were measured. The results firstly showed that FoxO1 interference increased the intracellular TG and lipids concentration (P < 0.05); and increased the proliferative index (PI), cell DNA synthesis, protein expression of Cyclin D1 in goose primary hepatocytes (P < 0.05). Secondly, the co-treatment of insulin and… More >

Abstract Goat milk is widely recognized for its nutritional value. Fatty acid synthase (FAS) is the crucial enzyme of fatty acid de novo synthesis. It plays an important role in the formation of goat milk fat. In this paper, we first introduced the molecular regulation process of goat milk fat metabolism based on the structure research of FAS. Secondly, we reviewed some key factors in FAS transcription and post-transcriptional regulation of the goat mammary gland and preliminarily constructed the expression network of the goat mammary gland FAS gene. The purpose of this paper is to systematically introduce More >

MicroRNA expression profile and lipid metabolism characteristics in liver of rat undergoing high-fat diet

    Wen JING1,§, Chuanfen LI2,3,§, Yingli LU4, Lianshi FENG4

    BIOCELL, Vol.43, No.3, pp. 129-138, 2019, DOI:10.32604/biocell.2019.06087

Abstract This study aimed to investigate the microRNA expression profile and the characteristics of lipid metabolism in the livers of rats undergoing a high-fat diet. Fifty male Sprague-Dawley (SD) rats were divided into a standard chow group (C group, N = 10) and a high-fat diet group (H group, N = 40). After 12 weeks, the rat body weight, body length, fat mass, and serum lipid concentration were measured. The expression profile of microRNAs and the gene and protein expression levels involved in lipid metabolism in rat liver were detected. Body fat and serum lipid… More >
