PIK3R1 as a Gastric Cancer Biomarker Linked to CD73+ Treg-Mediated Immunosuppression

    Bu Zou1,#, Yi-En Xu2,#, Hui-Chan He3, Zu-Lu Ye2, Da-Lei Zhou2, Cai-Yun He2,*, Chan Huang4,*

    Oncology Research, Vol.34, No.2, 2026, DOI:10.32604/or.2025.069453 - 19 January 2026

Abstract Objectives: Gastric cancer (GC) remains a major global health concern, and Phosphoinositide-3-Kinase Regulatory Subunit 1 (PIK3R1), a regulatory subunit of the PI3K signaling pathway, may play a critical yet underexplored role in GC progression. This study aimed to investigate the prognostic significance of PIK3R1 in GC and its association with the tumor immune microenvironment. Methods: PIK3R1 expression and its clinical relevance were analyzed using datasets from GC patients who underwent gastrectomy, including cohorts from The Cancer Genome Atlas (TCGA) and the Sun Yat-sen University Cancer Center (SYSUCC). Prognostic models integrating PIK3R1 expression with clinical parameters… More >

Characterization of Transcription Factor Krüppel-Like Factor 3 Expression in Splenic T Lymphocytes and Association with Immune Status in Septic Mice

    MIAO YAN1, DONGXUAN CHI2, WEN WANG3, PEI PEI4, MIN XIE1, SHUANGLING LI1,*

    BIOCELL, Vol.49, No.5, pp. 893-906, 2025, DOI:10.32604/biocell.2025.063622 - 27 May 2025

Abstract Background: Transcription factor Krüppel-like factor 3 (KLF3) may be involved in regulating inflammation and lymphocyte function. Immune dysfunction in sepsis involves both hyper-inflammation and immunosuppression. However, studies on T-lymphocyte KLF3 expression in sepsis are lacking. Methods: We induced sepsis in mice via cecal ligation and puncture (CLP), and their survival rate over 7 days was evaluated. To identify the immune status of these mice, we assessed their cytokine levels, organ damage scores, and splenic T-lymphocyte phenotype. Finally, T-lymphocyte KLF3 expression was detected through flow cytometry. Results: Over the 7 days of observation, septic mice demonstrated 64.7%… More >

Bladder squamous cell carcinoma in situ in the background of condyloma acuminatum in a kidney transplant recipient

    Crystal Lihong Yan1, Ryan A. Gallo2, Madhavi Pandiri3, Stephen J. Gallo4

    Canadian Journal of Urology, Vol.30, No.2, pp. 11505-11508, 2023

Abstract Condyloma acuminatum with synchronous squamous cell carcinoma in situ (CIS) rarely occurs in the bladder. In developed countries, bladder squamous cell carcinoma (SCC) is uncommon. Among the various noninvasive squamous bladder lesions, there is significant morphological overlap, which further complicates accurate diagnosis. Immunosuppression and human papilloma virus increase the risk of bladder condyloma acuminatum, which has a strong association with bladder SCC. Herein, we describe a case of a 79-year-old man with a history of end-stage renal disease with kidney transplantation and anal SCC who presented with bladder squamous cell CIS arising in the background More >

Scutellarin alleviates complete freund’s adjuvant-induced rheumatoid arthritis in mice by regulating the Keap1/Nrf2/HO-1 pathway

    JIAN LI1, QINGQING WANG2, XIAOYING ZHANG1,*

    BIOCELL, Vol.47, No.6, pp. 1307-1316, 2023, DOI:10.32604/biocell.2023.028714 - 19 May 2023

Abstract Scutellarin (SCU) is a herbal flavonoid glucuronide with multiple pharmacological activities, including anti-oxidant, anti-inflammation, vascular relaxation, anti-platelet, and myocardial protection. However, the effect of SCU on complete Freund’s adjuvant (CFA)-induced rheumatoid arthritis (RA) had not been studied. In this study, we investigated the beneficial effects of SCU in the CFA-induced RA mice model and the anti-arthritic activity was evaluated by paw edema. Enzyme-linked immunosorbent assay (ELISA) was carried out to evaluate the plasma levels of immunoglobulin (Ig)G, IgE, tumor necrosis factor (TNF)-α, interleukin (IL)-1β, IL-6, receptor activator of nuclear factor-κB ligand (RANKL), and osteoprotegerin (OPG).… More >

Knockdown of HVEM, a Lymphocyte Regulator Gene, in Ovarian Cancer Cells Increases Sensitivity to Activated T Cells

Ting Zhang1, Lei Ye1, Qizhi He, Jianlong Zhu

    Oncology Research, Vol.25, No.9, pp. 1665-1665, 2017, DOI:10.3727/096504017X15078984695565

Abstract Ovarian cancer is highly malignant with a gradually increasing incidence and a high mortality rate. Immunosuppression is induced in ovarian cancer, although the mechanism detail is not clear. It has been indicated that HVEM (herpesvirus entry mediator) B- and T-lymphocyte attenuator (BTLA) negatively regulates the immune responses of T lymphocytes. Here, HVEM mRNA was found to be elevated in ovarian cancer tissue samples and primary ovarian cancer cells in comparison with benign tissue samples. We then knocked down HVEM expression in an ovarian cancer cell line, OVCAR3, by lentivirus-based small hairpin RNA (shRNA). Cell Counting… More >

Suppression of interleukin-17 by type I interferons: a contributing factor in virus-induced immunosuppression?

Herbert Tilg, Alexander R. Moschen, Arthur Kaser

    European Cytokine Network, Vol.20, No.1, pp. 1-6, 2009, DOI:10.1684/ecn.2009.0141

Abstract Type I interferons (IFNs) are the first line of defence after various infections, and, as immuno-modulatory cytokines, bridge innate and adaptive immunity. IL-17, mainly secreted by specific T cells, has recently been identified regulating neutrophil-mediated inflammation, and has been implicated in the pathogen-esis of many acute and chronic inflammatory disorders. This cytokine is considered of critical importance for an effective anti-bacterial and anti-fungal immune response as needed subsequent to many viral infections. Recent studies have demonstrated that type I IFNs potently suppress IL-17 expression and Th17 differentiation in vitro and in vivo. Therefore, suppression of More >
