    BIOCELL, Vol.48, No.5, pp. 759-769, 2024, DOI:10.32604/biocell.2024.048551 - 06 May 2024

Abstract Background: Nonalcoholic fatty liver disease (NAFLD) is a global health concern with the acid sphingomyelinase (ASM)/ceramide (CE) pathway and the NOD-like receptor family, pyrin domain-containing protein 3 (NLRP3) inflammasome identified as pivotal players in lipid disorders and inflammation. This study explores the interaction mechanism between the ASM/CE pathway and NLRP3 in NAFLD cell models, aiming to understand the impact of amitriptyline (Ami), an ASM inhibitor, on lipid deposition and hepatocyte injury by regulating the ASM/CE-NLRP3 pathway. Methods: HepG2 and HL-7702 cells were exposed to free fatty acids (FFAs) to establish the NAFLD model. The cells… More >

Abstract In order to explore the role of forkhead box protein O1 (FoxO1) in the lipid metabolism and cell proliferation, goose primary hepatocytes were isolated and incubated with insulin or PI3K-Akt-mTOR pathway dual inhibitor NVP-BEZ235, and then transfected with FoxO1 interference plasmid. The related parameters of lipid metabolism and cell proliferation were measured. The results firstly showed that FoxO1 interference increased the intracellular TG and lipids concentration (P < 0.05); and increased the proliferative index (PI), cell DNA synthesis, protein expression of Cyclin D1 in goose primary hepatocytes (P < 0.05). Secondly, the co-treatment of insulin and… More >

Abstract According to the literature, different mechanisms and kinetics proceeding of regenerative growth has been established using the basic models of liver regeneration (after resection or chemically induced). Hence, in order to determine general regularities of the adaptive-compensatory processes in various pathological conditions, the processes taking place in the cholestatic liver of adult white rats during the first four days after common bile duct ligation have been studied. It has been shown that in cholestatic liver, compensatory-adaptive processes take place with different kinetics compared to those after resection. In particular, in response to the increased functional… More >

Plasma HGF and OPN as Potential Biomarkers of Pulmonary Arterial Hypertension in Congenital Heart Disease

    Dongdong Zheng1,#, Chi Shen1,2,#, Wenshi Liu1, Wenjing Lv1, Xiaofei Li1,*

    Congenital Heart Disease, Vol.16, No.4, pp. 373-381, 2021, DOI:10.32604/CHD.2021.015260 - 19 April 2021

Abstract Objectives: Pulmonary arterial hypertension in congenital heart disease (PAH-CHD) is the most common type of PAH and increases morbidity and mortality in patients with CHD. Right heart catheterization (RHC) is the standard method to diagnose PAH. However, RHC is an invasive and complicated method with relatively high cost. Noninvasive, feasible, and cost-efficient methods are urgently needed. The objective of this study was to evaluate three potential biomarkers of PAH-CHD: Hepatocyte growth factor (HGF), osteopontin (OPN), and suppression of tumorigenicity 2 (ST2). Methods: Plasma samples were collected from patients with CHD (n = 46) and healthy individuals (nMore >

Abstract Liver injury is often observed in various pathological conditions including posthepatectomy state and cancer chemotherapy. It occurs mainly as a consequence of the combined necrotic and apoptotic types of cell death. In order to study liver/hepatocyte injury by the necrotic type of cell death, we studied signal-regulated necrosis (necroptosis) by developing a new optic probe for detecting receptor-interacting protein kinase 1 (RIP)/RIP3 binding, an essential process for necroptosis induction. In the mouse hepatocyte cell line, TIB-73 cells, TNF-a/cycloheximide (T/C) induced RIP1/3 binding only when caspase activity was suppressed by the caspase-specific inhibitor z-VAD-fmk (zVAD). T/C/zVAD-induced… More >

    Oncology Research, Vol.25, No.7, pp. 1039-1046, 2017, DOI:10.3727/096504016X14813914187138

Abstract Hepatocyte cell adhesion molecule (hepaCAM), a new type of CAM, belongs to the immunoglobulin superfamily. Recently, hepaCAM was reported to be implicated in cancer development, and many researchers investigated its biological function in the tumorigenesis of various cancers. However, what kind of role hepaCAM plays in colorectal cancer (CRC) remains unknown. In this study, we found that hepaCAM was downregulated in CRC tissues and cell lines. Overexpression of hepaCAM inhibited CRC cell proliferation, migration, and invasion in vitro. Furthermore, the tumorigenesis assay showed that increased expression of hepaCAM suppressed CRC tumor growth and metastasis in More >

Abstract Low pressure plasma polymer films were synthesized using pyrrole and allylamine monomers and adding iodine was used (or not) for the reaction in both cases. They were polymerized on glass substrates under the same reaction conditions. Polymerization of allylamine was also studied at different operating powers. These thin polymer films were used as culture surfaces for HepG2 cells, a cell line derived from a human hepatoma. The proliferation, differentiation and two-dimensional propagation until obtaining monolayer of the cells was studied on the different synthetized films and correlations were established between the conditions of synthesis, the More >

    BIOCELL, Vol.39, No.2-3, pp. 9-14, 2015, DOI:10.32604/biocell.2015.39.009

Abstract We have designed and evaluated a radial-flow bioreactor for three-dimensional liver carcinoma cell culture on a new porous coated scaffold. We designed a culture chamber where a radial flow of culture medium was continuously delivered through it. Once this system was established, flow was simulated using flow dynamics software based on numeric methods to solve Navier-Stockes flow equations. Perfusion within cell culture scaffolds was simulated using a flow velocity of 7 mL/min and found that cell culture medium was distributed unhindered in the bioreactor chamber. Afterwards, the bioreactor was built according to the simulated design More >

Alanine minimises hepatocyte injury after ischemia-reperfusion in an ex vivo rat liver model

    Berengere PAPEGAY1, *, Michaela STADLER1, Vincent Nuyens1, Isabelle SALMON2, Veronique KRUYS3, Jean G. BOOGAERTS1

    BIOCELL, Vol.38, No.1, pp. 25-32, 2014, DOI:10.32604/biocell.2014.38.025

Abstract Ischemia-reperfusion injury is a determinant in liver injury occurring during surgery, ischemic states and multiple organ failure. The pre-existing nutritional status of the liver, i.e., fasting, might contribute to the extent of tissue injury. This study investigated whether alanine, an amino acid precursor of glucose, could protect ex vivo perfused livers of fasting rats from reperfusion injury. The portal vein was cannulated, the liver removed and perfused in a closed ex vivo system. Isolated livers were perfused either with glucose 1 g/L and 10 g/L, or with equal concentrations of alanine (n = 10 in… More >
