Abstract Background: ((1-triphenylaminebenzo[c][1,2,5] thiadiazole-4-yl) styryl)-1-methylpyridin methylpyridin-1-ium iodide salt (TBZPy) is a novel photosensitizer that displays excellent photodynamic properties. However, There are few reports on the mechanism of action of the TBZPy photodynamic. Previous studies revealed that photodynamic therapy (PDT) could induce endoplasmic reticulum stress by acting on the endoplasmic reticulum. Therefore, in this study, we investigated the effects of endoplasmic reticulum stress induced by TBZPy-PDT in treating High-risk human papillomavirus (HR-HPV) infection and their underlying mechanisms. Methods: The human cervical cancer cell line HeLa (containing whole genome of HR-HPV18) was treated with TBZPy-PDT. Cell migration, invasion,… More > Graphic Abstract

Raloxifene-loaded and aptamer-bonded exosomes induce autophagic and apoptotic death in HeLa cells by enhancing the lysosomotropic effect

    OMER ERDOGAN1, GULEN MELIKE DEMIRBOLAT2, OZGE CEVIK1,*

    BIOCELL, Vol.47, No.5, pp. 1051-1063, 2023, DOI:10.32604/biocell.2023.028129 - 10 April 2023

Abstract Background: Raloxifene, a selective estrogen receptor modulator, is also known to be a lysosomotropic agent. The bioavailability of raloxifene is around 2% due to extensive hepatic transport. Exosomes are nanosized vesicles that are naturally released from cells. Method: In this study, exosomes released from HeLa cervical cancer cells were loaded with raloxifene to increase its bioavailability, and an aptamer was attached to the exosome membrane for targeting only HeLa cells. Characterization of exosomes isolated from HeLa cells was performed by transmission electron microscopy, zeta sizer, and western blotting. In addition, the cytotoxic, apoptotic, autophagic, and lysosomotropic More >

    Oncologie, Vol.23, No.2, pp. 279-291, 2021, DOI:10.32604/Oncologie.2021.015969 - 22 June 2021

Abstract Spheroids are generally self-assembled cells with the ability to generate their extracellular matrix, including the complex cell-matrix and the cell-cell interactions that resemble the functional characteristics of the corresponding tissue in vivo. The study aimed to develop a three-dimensional (3D) spheroid system for the cervical cancer cell lines, HeLa (HPV18), CaSki (HPV16), SiHa (HPV16), C33A (non-HPV), HT3 (non-HPV) as well as to identify its biological activity in the extracellular form. For the formation of the cervical cancer spheroids, the liquid overlay approach was applied, followed by embedding to the bovine collagen I matrix. Spheroid formation using More >

Abstract While phthalate acid esters (PAEs) cannot fluoresce alone, they can be detected by fluorescence spectroscopy after chelation with bovine serum albumin (BSA). In this study, the types of amino acid residues at the active site of PAEs chelated with BSA were determined using molecular docking technology. A modification scheme of BSA with higher detection sensitivity fluorescence spectroscopy for PAEs was proposed based on the docking results and constructed for a novel BSA structure with a higher detection sensitivity of fluorescence spectroscopy using a homologous modeling method. Density functional theory (DFT) was employed to explore the… More >

Overexpression of inhibin α (1-32) fusion protein promotes apoptosis and cell cycle arrest in a cervical cancer cell model (Hela cells)

    BIOCELL, Vol.38, No.1, pp. 17-24, 2014, DOI:10.32604/biocell.2014.38.017

Abstract Inhibins play important roles in the reproductive system. To evaluate whether inhibin α (1-32) fusion protein plays a role in cervical cancer growth, the plasmid pVAX-inhα was constructed and its effect on proliferation and apoptosis of the human cervical cancer cell line (Hela) was checked by flow cytometry and real-time PCR. The expression and localization of inhibin α protein were detected by RT-PCR and confocal microscopy which showed that inhibin α protein was expressed and localized in the nucleus of Hela cells. Over expression of inhibin α gene significantly induced cell apoptosis and ceased S… More >

    BIOCELL, Vol.27, No.3, pp. 301-309, 2003, DOI:10.32604/biocell.2003.27.301

Abstract The effects of Photodynamic Therapy using 2nd generation photosensitizers have been widely investigated aiming clinical application treatment of solid neoplasms. In this work, ultrastructure changes caused by the action of two 2nd generation photosensitizers and laser irradiation on CHO-K1 and HeLa (neoplastic) cells were analyzed by transmission electron microscopy. Aluminum phthalocyanine chloride, aluminum phthalocyanine tetrasulfonate chloride and radiation from a semiconductor laser at a fluency of 0.5 J/ cm2 (Power=26mW; λ=670nm) were used. The results showed induction of apoptosis. Such alterations where observed in HeLa but not in CHO-K1 cells after Aluminum phthalocyanine tetrasulfonate chloride (AlPcS4) photodynamic More >
