Abstract Introduction: Transplantation of mesenchymal stromal cells (MSCs) is a promising therapy for type 1 diabetes (T1D). However, whether the infused MSCs affect the endoplasmic reticulum stress or subsequent unfolded protein response in β cells remains unclear. Methods: To investigate this, we induced early-onset T1D in non-obese diabetic mice using streptozotocin. Subsequently, T1D mice were randomly assigned to receive either MSCs or phosphate-buffered saline. We observed the in vivo homing of MSCs and assessed their effectiveness by analyzing blood glucose levels, body weight, histopathology, pancreatic protein expression, and serum levels of cytokines, proinsulin, and C-peptide. Results: Infused MSCs… More > Graphic Abstract

Abstract Background: ((1-triphenylaminebenzo[c][1,2,5] thiadiazole-4-yl) styryl)-1-methylpyridin methylpyridin-1-ium iodide salt (TBZPy) is a novel photosensitizer that displays excellent photodynamic properties. However, There are few reports on the mechanism of action of the TBZPy photodynamic. Previous studies revealed that photodynamic therapy (PDT) could induce endoplasmic reticulum stress by acting on the endoplasmic reticulum. Therefore, in this study, we investigated the effects of endoplasmic reticulum stress induced by TBZPy-PDT in treating High-risk human papillomavirus (HR-HPV) infection and their underlying mechanisms. Methods: The human cervical cancer cell line HeLa (containing whole genome of HR-HPV18) was treated with TBZPy-PDT. Cell migration, invasion,… More > Graphic Abstract

Abstract Background: While the Bushen Yizhi Formula can treat Alzheimer’s disease (AD), the yet to be ascertained specific mechanism of action was explored in this work. Methods: Different concentrations of the Bushen Yizhi Formula and amyloid-beta peptide (Aβ) were used to treat rat pheochromocytoma cells (P12) and human neuroblastoma cells (SH-SY5Y). Cell morphological changes were observed to determine the in vitro cell damage. Cell Counting Kit (CCK)-8 assay and flow cytometry were employed to identify cell viability and apoptosis/cell cycle, respectively. Western blotting and immunohistochemistry were employed to measure the expressions of endoplasmic reticulum stress (ERS)-related proteins (GRP78… More >

Konjac glucomannan enhances 5-FU-induced cytotoxicity of hepatocellular carcinoma cells via TLR4/PERK/CHOP signaling to induce endoplasmic reticulum stress

YONGKANG SHI, JUN MA, KE CHEN, BIN CHEN*

    Oncology Research, Vol.30, No.4, pp. 201-210, 2022, DOI:10.32604/or.2022.027584 - 31 January 2023

Abstract 5-Fluorouracil (5-FU) is a commonly used chemotherapeutic agent for various cancers. However, the drug resistance developed by tumor cells hinders the therapeutic effect. Konjac glucomannan (KGM) is indicated to sensitize 5-FU-resistant hepatocellular carcinoma (HCC) cells to 5-FU. In our study, we found that KGM or 5-FU treatment alone did not affect the malignant cell behaviors and endoplasmic reticulum (ER) stress of 5-FU-resistant HCC cells or HepG2/5-FU and Bel-7402/5-FU cells, while cotreatment with KGM and 5-FU significantly facilitated HCC cell apoptosis and ER stress and suppressed cell proliferation potential and migration abilities. Moreover, we explored the… More >

Abstract Spinal cord injury (SCI) remains an intractable clinical challenge of neurosurgery, it can be divided into two stages: uncontrollable primary injury induced by mechanical damage and controllable secondary injury regulated by continuous cell death. The apoptosis was the one of most important events in secondary injury, previous studies revealed that excessive endoplasmic reticulum (ER) stress breaks down the homeostasis and triggers apoptosis in the spinal cord. To deter or alleviate the secondary jury, we screen one of fat-soluble compounds, salubrinal, which was an inhibitor of eIF2α dephosphorylation can repair SCI by inhibiting ER stress in… More >

Nicotinic acid induces apoptosis of glioma cells via the calcium-dependent endoplasmic reticulum stress pathway

    XIANGCAI YANG1, JIAGUI QU2, JIEJING LI1,*

    BIOCELL, Vol.46, No.4, pp. 1041-1051, 2022, DOI:10.32604/biocell.2022.017383 - 15 December 2021

Abstract Malignant glioma is one of the most common and deadly tumors in the central nervous system while developing effective treatments for this devastating disease remains a challenge. Previously, we demonstrated that the vitamin nicotinic acid (NA) inhibits glioma invasion. Here, we show that high-dose NA induces apoptosis of malignant glioma cells in vitro and in vivo. In cultured U251 glioma cells treated with NA, we detected ER stress that was likely caused by elevated intracellular calcium levels. The elevated calcium can be attributed to the activation of TRPV1, a cation channel that has been implicated in cutaneous More >

Abstract Zinc is a trace element involved in maintaining cellular structure and function. Although zinc is associated with left ventricular hypertrophy (LVH), there have been few reports on this association. This study aimed to evaluate the correlation between Zip14 and expression of endoplasmic reticulum stress (ERS) associated molecules in hypertrophied hearts of rats. Dahl salt-sensitive rats were fed a high salt diet to establish a left ventricular hypertrophy (LVH) rat model. RT-PCR was used to determine Zip14, activating transcription factor (ATF4), ATF6, x-box-binding protein 1 (xBP1), C/EBP homologous protein (CHOP), immunoglobulin-binding protein (BiP) mRNA expression. Nelfinavir and Ritonavir Kill Bladder Cancer Cells Synergistically by Inducing Endoplasmic Reticulum Stress

Akinori Sato, Takako Asano, Kazuki Okubo, Makoto Isono, Tomohiko Asano

    Oncology Research, Vol.26, No.2, pp. 323-332, 2018, DOI:10.3727/096504017X14957929842972

Abstract The human immunodeficiency virus (HIV) protease inhibitor nelfinavir acts against malignancies by inducing endoplasmic reticulum (ER) stress. The HIV protease inhibitor ritonavir, on the other hand, not only induces ER stress but also inhibits P-glycoprotein’s pump activity and thereby enhances the effects of its substrate drugs. We therefore postulated that ritonavir in combination with nelfinavir would kill bladder cancer cells effectively by inducing ER stress cooperatively and also enhancing nelfinavir’s effect. Nelfinavir was shown to be a P-glycoprotein substrate, and the combination of nelfinavir and ritonavir inhibited bladder cancer cell growth synergistically. It also suppressed… More >

Abstract Previous studies have shown that 5,2',4'-trihydroxy-6,7,5'-trimethoxyflavone (TTF1) is the primary anticancer constituent of the traditional Chinese medicinal plant Sorbaria sorbifolia (SS), which has been applied to treat cancer in China. In this study, we investigated the in vitro and in vivo antitumor effects and biological mechanisms of small-molecule TTF1 nanoparticles (TTF1-NPs). The effects of TTF1-NPs on cell growth and apoptosis were investigated using human hepatoma cells. The molecular changes associated with the effects of TTF1-NPs were analyzed by immunocytochemistry and Western blot analysis. The in vivo effect of TTF1-NPs was investigated using the HepG2 tumor xenograft More >
