    PD-1+ and TIM-3+ T cells widely express common γ-chain cytokine receptors in multiple myeloma patients, and IL-2, IL-7, IL-15 stimulation up-regulates PD-1 and TIM-3 on T cells

    Oncology Research, Vol.32, No.10, pp. 1575-1587, 2024, DOI:10.32604/or.2024.047893 - 18 September 2024

Abstract Background: Immune checkpoint ligand-receptor interactions appear to be associated with multiple myeloma (MM) progression. Simultaneously, previous studies showed the possibility of PD-1 and TIM-3 expression on T cells upon stimulation with common γ-chain family cytokines in vitro and during homeostatic proliferation. The aim of the present work was to study the impact of homeostatic proliferation on the expansion of certain T cell subsets up-regulating PD-1 and TIM-3 checkpoint molecules. Methods: The expression of CD25, CD122, CD127 common γ-chain cytokine receptors, phosphorylated signal transducer and activator of transcription-5 (pSTAT5) and eomesodermin (EOMES) was comparatively assessed with flow… More >

Calcitonin gene related peptide modified mesenchymal stem cells reduce restenosis after carotid balloon injury in rats

    PANKE CHEN*, SHUAI MA

    BIOCELL, Vol.46, No.8, pp. 1903-1909, 2022, DOI:10.32604/biocell.2022.018651 - 22 April 2022

Abstract This work aimed to investigate the effects of calcitonin gene-related peptide (CGRP)-modified mesenchymal stem cells (MSCs) on vascular stenosis in carotid balloon-injured rats. The CGRP gene labeled with EGFP was transfected into bone marrow MSCs, and CGRP protein expression in MSCs was confirmed by immunofluorescence assays. A rat carotid balloon injury model was established using a surgical method. CGRP-modified MSCs were orthotopically transplanted into the injured area of the rats. At 28 days after cell transplantation, EGFP and CD31 expression was detected by immunofluorescence staining. Hematoxylin-eosin (H&E) staining was used to detect the intima/media area… More >

Transplanted choroidal plexus epithelial cells can integrate with organotypic spinal cord slices into a new system

    JINGJIE LIU1,#, XIAOYAN DING2,#, LI XIANG1, SHENGLI HUANG3,*

    BIOCELL, Vol.46, No.6, pp. 1537-1544, 2022, DOI:10.32604/biocell.2022.018441 - 07 February 2022

Abstract This study aimed to evaluate the integration of transplanted choroidal plexus epithelial cells with organotypic spinal cord slices. Organotypic spinal cord slices, normally cultured for 6 days, were divided into control group (Ctrl) and transplanted group (T). The choroidal plexus epithelial cells were dissociated and primary cultured (C group). The choroidal plexus epithelial cells cultured for 6–7 days were labeled by 1,1’-dioctadecyl-3,3,3’,3’-tetramethyl-indocarbocyanineperchlorate (CM-Dil), and were identified by transthyretin (TTR) in immunocytochemistry. They were adjusted to the density of 0.5–1 × 107/ml, then 2 μl cells suspension were transplanted to the spinal cord slices in the T… More >

Age-related modifications of macrophages influenced by “inflammageing” in graft vs. host disease

    YAQUN HONG1,2, BO WAN3, XIAOFAN LI1,4,*

    BIOCELL, Vol.44, No.2, pp. 237-246, 2020, DOI:10.32604/biocell.2020.08887 - 27 May 2020

Abstract Most studies focus on the adaptive immune cells in the GVHD pathogenesis, while little is known about innate immune cells in GVHD occurrence and development, especially macrophages. Meanwhile, a higher incidence of graft versus host disease (GVHD) is also found in the elderly patients. Though advances have been made in the modification of macrophages influenced by the inflamm-ageing, there is still no review on the role of macrophages in GVHD and the association between GVHD and the altered macrophages by inflamm-ageing. In this review, we focus on the potential age-related modifications of macrophage in GVHD, More >
