Abstract Bone metastasis secondary to breast cancer negatively impacts patient quality of life and survival. The treatment of bone metastases is challenging since many anticancer drugs are not effectively delivered to the bone to exert a therapeutic effect. To improve the treatment efficacy, we developed Pluronic P123 (P123)-based polymeric micelles dually decorated with alendronate (ALN) and cancer-specific phage protein DMPGTVLP (DP-8) for targeted drug delivery to breast cancer bone metastases. Doxorubicin (DOX) was selected as the anticancer drug and was encapsulated into the hydrophobic core of the micelles with a high drug loading capacity (3.44%). The… More > Graphic Abstract

    Oncology Research, Vol.32, No.1, pp. 95-98, 2024, DOI:10.32604/or.2023.046174 - 15 November 2023

Abstract Greenblatt and his team have unveiled vertebral skeletal stem cells (vSSCs) as a critical player in the landscape of bone metastasis. This commentary delves into the transformative discoveries surrounding vSSCs, emphasizing their distinct role in bone metastasis compared to other stem cell lineages. We illuminate the unique properties and functions of vSSCs, which may account for the elevated susceptibility of vertebral bones to metastatic invasion. Furthermore, we explore the exciting therapeutic horizons opened by this newfound understanding. These include potential interventions targeting vSSCs, modulation of associated signaling pathways, and broader implications for the treatment and More > Graphic Abstract

FRANCISCO RAÚL BORZONE1,*, MARÍA BELÉN GIORELLO1, LEANDRO MARCELO MARTINEZ2, MARÍA CECILIA SANMARTIN1,3, LEONARDO FELDMAN4, FEDERICO DIMASE5, EMILIO BATAGELJ6, GUSTAVO YANNARELLI3, NORMA ALEJANDRA CHASSEING1,*

    Oncology Research, Vol.31, No.3, pp. 361-374, 2023, DOI:10.32604/or.2023.028104 - 22 May 2023

Abstract Breast cancer is the predominant form of carcinoma among women worldwide, with 70% of advanced patients developing bone metastases, with a high mortality rate. In this sense, the bone marrow (BM) mesenchymal stem/stromal cells (MSCs) are critical for BM/bone homeostasis, and failures in their functionality, transform the BM into a pre-metastatic niche (PMN). We previously found that BM-MSCs from advanced breast cancer patients (BCPs, infiltrative ductal carcinoma, stage III-B) have an abnormal profile. This work aims to study some of the metabolic and molecular mechanisms underlying MSCs shift from a normal to an abnormal profile… More >

    Oncologie, Vol.24, No.3, pp. 599-604, 2022, DOI:10.32604/oncologie.2022.023310 - 19 September 2022

Abstract Gastric cancer is the fifth most commonly diagnosed cancer and the fourth leading cause of cancer death worldwide in 2020. Gastric cancer usually undergoes lymph node metastasis and implantation metastasis, but bone metastasis and bone marrow invasion are rare. However, gastric cancer patients with bone marrow invasion usually have cancer emergency, so special attention should be paid in clinical practice. Herein, we analyzed the clinical characteristics of an asian gastric cancer patient with bone marrow invasion and disseminated intravascular coagulation (DIC) in our hospital and summarized the diagnosis and treatment experience to provide a reference More >

A need for stratification of metastasis samples according to secondary site in gene expression studies

    IVANA SAMARŽIJA1,2,*

    BIOCELL, Vol.46, No.7, pp. 1747-1750, 2022, DOI:10.32604/biocell.2022.018784 - 17 March 2022

Abstract Comparisons of gene expression profiles between primary tumors and metastasis have revealed genes that are implicated in metastasis formation. However, gene expression studies conducted on metastasis samples from the same primary site usually do not discriminate between different secondary sites. Although the change in the expression of number of genes is expected to be common to metastasis from the same primary but different secondary sites, herein the data that point to substantial differences are presented. Furthermore, the reciprocal communication between metastatic and host cells that is influencing these differences is outlined to emphasize the need More >

SPAG9 promotes prostate cancer growth and metastasis

    Chunhua YANG1,2,3, Ye TIAN1,2,3

    BIOCELL, Vol.43, No.3, pp. 207-214, 2019, DOI:10.32604/biocell.2019.07258

Abstract Sperm-associated antigen 9 (SPAG9) expression is increased in prostate tissues of prostate cancer patients. This experimental study aimed to investigate the role of SPAG9 in bone metastasis of prostate cancer. Immunohistochemical analysis showed that SPAG9 staining was positive in 81.67% of 240 cases of prostatic carcinoma but only in 6.67% of 120 cases of benign prostate hyperplasia. Strong PAG9 staining was positively correlated with Gleason score and bone metastasis in 240 prostate cancer patients (p < 0.05), but not with the age or serum prostatespecific antigen level (p > 0.05). PC-3 cells were transfected with shRNA… More >

Abstract Osteoprotegerin (OPG) is a robust antiresorptive molecule that acts as a decoy receptor for the receptor activator of nuclear factor kB ligand (RANKL), the mediator of osteoclastogenesis. This study was designed to explore the possible role of serum OPG and RANKL in detecting bone metastasis in breast cancer and its interaction with clinicopathologic parameters. Serum levels of RANKL and OPG were estimated in 44 metastatic and 36 nonmetastatic breast cancer patients using ELISA kits. Serum OPG levels were significantly reduced in patients with bone metastasis and correlated negatively with the number of bone lesions and… More >

Overexpression of Interferon Regulatory Factor 7 (IRF7) Reduces Bone Metastasis of Prostate Cancer Cells in Mice

Yang Zhao, Wenxia Chen, Weiliang Zhu, Hui Meng, Jie Chen, Jian Zhang

    Oncology Research, Vol.25, No.4, pp. 511-522, 2017, DOI:10.3727/096504016X14756226781802

Abstract The purpose of this study was to identify the role of interferon regulatory factor 7 (IRF7) in the bone metastasis of prostate cancer. Herein we demonstrated the lower expression of IRF7 in bone metastases of prostate cancer. Overexpression of IRF7 in prostate cancer cells had a marked effect on inhibiting bone metastases but not on tumor growth in xenograft nude mice. While in vitro, upregulation of IRF7 had little effect on the malignant phenotype of prostate cancer cells including proliferation, apoptosis, migration, and invasion. However, prostate cancer cells overexpressing IRF7 significantly enhanced the activity of More >

Hailin Pang1, Ningqiang Ma1, Mi Jiao, Weiwei Shen, Bo Xin, Tongfei Wang, Feng Zhang, Lili Liu, Helong Zhang

    Oncology Research, Vol.25, No.1, pp. 35-42, 2017, DOI:10.3727/096504016X14719078133249

Abstract The bone is among the most common sites of metastasis in patients with lung cancer. Over 30%–40% of lung cancers can develop bone metastasis, and no effective therapeutic methods exist in clinic cases. Wnt/β-catenin signaling and Dickkopf1 (DKK1) play important roles in the progression of lung cancer, which preferentially metastasizes to the skeleton. However, the role of DKK1 in osteotropism of small cell lung cancer (SCLC) remains to be elucidated. This study aimed to define the role of DKK1 in SCLC bone metastasis and investigate the underlying mechanisms. Our results demonstrated that the expression level… More >
